Insights into Assessing the Genetics of Endometriosis
This review summarizes recent genome-wide studies investigating genetic variants contributing to endometriosis and discusses future research directions.

This review examines how genetic susceptibility to endometriosis has been studied from 2008–2012, comparing hypothesis-driven candidate gene association studies with newer hypothesis-free genome-wide approaches, and summarizing evidence from published genetic association work. It highlights that candidate gene studies tested 73 genes using SNP-based association designs, but that no candidate gene had been robustly associated with endometriosis across studies, with VEGF showing only suggestive findings that failed to replicate in larger datasets. The paper emphasizes key limitations for gene-discovery efforts, including problems with replication, differences in study design and disease definitions, low power due to small effect sizes, and issues such as winner’s curse and varying linkage disequilibrium across ethnic populations.
