The study evaluated two anti–TNF-α inhibitors, recombinant human TNF receptor: Fc fusion protein (rhTNFR: Fc) and TNF-α monoclonal antibody, in human eutopic endometrial stromal cells (hESCs), comparing effects on cell survival, cell cycle, and invasiveness following TNF-α stimulation. rhTNFR: Fc suppressed TNF-α–induced hESC survival and invasiveness, and reduced the proportion of cells in S phase, whereas the TNF-α monoclonal antibody did not produce these effects; a caveat is that the in vitro component was limited to hESCs and directly tested only these two TNF-α inhibitor formats. The authors then used a surgically induced mouse model of endometriosis and found that rhTNFR: Fc treatment decreased fluorescence intensity and the size of implanted endometriotic lesions. This paper is centrally about endometriosis — it tests rhTNFR: Fc as an inhibitor of lesion development and links its effects to reduced hESC survival, proliferation (S-phase reduction), and invasiveness.
