Dose–response effect of interleukin (IL)-1β, tumour necrosis factor (TNF)-α, and interferon-γ on the in vitro production of epithelial neutrophil activating peptide-78 (ENA-78), IL-8, and IL-6 by human endometrial stromal cells
This study analyzed the dose-dependent production of ENA-78, IL-6, and IL-8 by human endometrial stromal cells stimulated by IL-1β, TNF-α, and interferon-γ.

The study examined how different doses of IL-1β, TNF-α, and interferon-γ affect in vitro production of the neutrophil-activating chemokine ENA-78 and the cytokines IL-8 and IL-6 by human endometrial stromal cells. Stromal cells derived from eutopic endometrium of seven cycling endometriosis-free women were isolated and cultured (including monolayers) and then exposed to IL-1β (0.08–50 ng/mL), TNF-α, and interferon-γ (20–500 ng/mL), with ENA-78, IL-8, and IL-6 release measured. IL-1β increased all three mediators dose-dependently but showed decreased IL-8 and IL-6 release at the highest IL-1β dose tested, while TNF-α produced a plateau effect across a dose range and interferon-γ had opposite effects by inhibiting IL-8 above 20 ng/mL while stimulating IL-6 and largely leaving ENA-78 unaffected. Limitations include small sample size (n=7) and reliance on eutopic, endometriosis-free stromal cells rather than endometriotic tissue. This paper is centrally about endometriosis in its use of IL-8 and ENA-78 biology, explicitly noting that IL-8 and ENA-78 are increased in endometriosis peritoneal fluid and framing stromal cytokine regulation relevant to endometriosis.
