Role of Estrogen Receptor-β in Endometriosis
This review examines how elevated estrogen receptor-β, caused by promoter hypomethylation, drives endometriosis progression by suppressing ERα and promoting inflammation and progesterone resistance.

The paper reviews and presents experimental work on how nuclear estrogen receptor biology, especially estrogen receptor-β (ERβ), differs between endometriotic tissue and eutopic endometrium, focusing on stromal cells from ovarian endometriosis. Using prior and newly discussed RT-PCR comparisons, methylation assays, and ERβ knockdown/overexpression experiments in cultured primary stromal cells, it reports markedly higher ERβ with lower ERα and progesterone receptor (PR) levels in endometriosis, and identifies hypomethylation of a CpG island at the ERβ promoter as a primary mechanism driving this differential expression; it also shows ERβ can suppress ERα promoter activity. A stated limitation is the reliance on cultured primary stromal cell models (and inferences about in vivo regulation from related observations) rather than direct in vivo mechanistic testing for every step.
