Endometriosis patients showed elevated peritoneal fluid cytokines IL-1beta, IL-6, and IGF-1, and this fluid enhanced endothelial cell proliferation, indicating angiogenic potential that may support endometrial transplant growth.

The study measured concentrations of proinflammatory cytokines (IL-1β, IL-6) and angiogenic/growth factors (vascular endothelial growth factor, tumor growth factor-β, and insulin-like growth factor-1) in peritoneal fluid from patients with external genital endometriosis and healthy women, and assessed how patient peritoneal fluid affected proliferative activity of EA.Hy926 human endothelial cells using flow-cytometry cell-cycle analysis. Peritoneal fluid from patients showed increased IL-1β, IL-6, and insulin-like growth factor-1 compared with controls, and significantly increased endothelial mitotic activity and angiogenic potential, whereas healthy controls did not. A stated caveat is that angiogenic effects were evaluated in an in vitro endothelial cell model rather than directly in clinical tissue or implantation settings. This paper is centrally about endometriosis — it links peritoneal cytokine profiles and endothelial angiogenic potential to external genital endometriosis.

Abstract

The concentrations of proinflammatory cytokines (IL-1beta, IL-6), vascular endothelium growth factor, tumor growth factor-beta, and insulin-like growth factor-1 were measured in the peritoneal fluid of patients with external genital endometriosis and healthy women by enzyme immunoassay. The effect of peritoneal fluid from patients with external genital endometriosis on proliferative activity of EA.Hy926 human endothelial cells was evaluated by the method based on the analysis of cell cycle by flow cytometry. The concentrations of IL-1beta, IL-6, and insulin-like growth factor-1 were increased in patients with endometriosis in comparison with healthy women. The peritoneal fluid from patients with endometriosis (but not from healthy women) significantly increased mitotic activity of endothelial cells and exhibited high angiogenic potential, which can promote implantation and growth of endometrial transplants. Presumably, insulin-like growth factor-1 stimulates this process.
