Epigenetic alterations of CYP19A1 gene in Cumulus cells and its relevance to infertility in endometriosis
Endometriosis patients with infertility showed lower CYP19A1 gene expression in cumulus cells due to epigenetic alterations including DNA methylation, histone modifications, and altered ERβ binding.

This study examined epigenetic mechanisms underlying aberrant aromatase (CYP19A1) expression in cumulus cells from 24 infertile women undergoing ovarian stimulation for ICSI, comparing patients with and without endometriosis. CYP19A1 mRNA levels were measured by RT-qPCR, while DNA methylation, histone modifications, and ERβ binding to CYP19A1 regulatory promoters were assessed using ChIP. Endometriosis patients showed significantly lower CYP19A1 expression, alongside increased MeCP2 incorporation and changes in histone marks (including hypoacetylation at H3K9 on PII and hypermethylation at H3K9 on PI.4), and differential ERβ binding across CYP19A1 promoters. The paper does not provide additional explicit caveats in the provided text beyond interpreting these epigenetic alterations as potentially impairing follicular steroidogenesis and oocyte/embryo condition. This paper is centrally about endometriosis—specifically epigenetic regulation of CYP19A1 (aromatase) in cumulus cells as a contributor to infertility in endometriosis.
