The study investigated whether the synthetic retinoid fenretinide, which promotes apoptosis, could target endometriotic stromal cells by restoring retinoid signaling; primary human endometriotic stromal cells were treated with fenretinide across 0–20 µmol/L and assessed for viability, apoptosis markers (including PARP), proliferation (PCNA), and expression of genes involved in retinoid uptake/action (STRA6, CRABP2, FABP5). Fenretinide reduced total cell count and viability at higher doses, increased PARP as an apoptotic marker, decreased PCNA after prolonged treatment, and induced STRA6 expression while CRABP2 and FABP5 were unchanged, in line with altered retinoid availability/action; in a mouse xenograft model, fenretinide treatment for 2 weeks significantly decreased endometriotic lesion volume. The paper’s caveats include reliance on in vitro primary cell cultures and a subcutaneous xenograft model, which may not fully replicate lesion microenvironments.
