The study tested whether TRIzol extraction from secretory-phase endometrial biopsies enables combined mRNA and protein profiling from the same sample in women without endometriosis (controls, n=6) and with minimal-to-mild (stages I–II, n=5) or moderate-to-severe (stages III–IV, n=5). Using SELDI-TOF mass spectrometry on the TRIzol protein fraction, the authors found 32 differentially expressed mass peaks across all endometriosis stages versus controls, while previously reported mRNA microarray differences in the same samples involved 9 representative genes. A major limitation explicitly noted is that no corresponding proteins/genes were found between the transcriptomic and proteomic results, which the authors attribute to factors including differences in protein size range captured by SELDI-TOF and potential disconnect between transcript and protein activity due to post-translational modifications.
