Nuclear Receptor, Coregulator Signaling, and Chromatin Remodeling Pathways Suggest Involvement of the Epigenome in the Steroid Hormone Response of Endometrium and Abnormalities in Endometriosis

This study examined how nuclear receptors and their coregulators, along with linked steroid-hormone signaling pathways, regulate gene expression in human endometrium across the menstrual cycle and in women with endometriosis. Using polymerase chain reaction arrays profiling 84 genes and immunohistochemical validation of select targets, the authors reported that DNA methylation and transcriptional repression signaling were the most affected pathways in endometrium from women with versus without endometriosis, independent of cycle phase. They also found that thyroid hormone receptor and vitamin D receptor pathways were regulated in both normal cycling endometrium and disease endometrium via changes in vitamin D– or thyroid hormone–regulated genes. A limitation explicitly implied by the approach is that gene expression was assessed using a restricted set of genes and pathway inference from array data. This paper is centrally about endometriosis — it analyzes nuclear receptor/coregulator signaling and epigenome-linked pathways in endometrium in women with endometriosis.
