Endometriosis, endocrine disrupters, and epigenetics: an investigation into the complex interplay in women with polybrominated biphenyl exposure and endometriosis

This cross-sectional study examined DNA methylation differences in peripheral blood from 305 women in the Michigan polybrominated biphenyl (PBB) Registry, using the MethylationEPIC BeadChip and linear regression of each CpG site with endometriosis status as the outcome, with sensitivity analyses controlling for estradiol levels and menopausal status. The authors report that 39,877 CpG sites were nominally associated with endometriosis after adjusting for age and cellular heterogeneity, and pathway enrichment implicated endocrine, immunologic, oncologic, and cell-regulation processes, including FoxO, Wnt, and Hedgehog signaling. However, no CpGs remained significant after multiple-comparison correction (FDR < 0.05), limiting statistical support for specific methylation differences.
