The study established a novel human endometrial mesenchymal stem cell (eMSC) line from a patient with adenomyosis and compared its properties with eMSCs derived from a healthy donor. The patient- and donor-derived cells both showed fibroblast-like morphology, had similar expression of surface markers, and did not differ in adipogenic potential, while karyotype analysis at passages 6–7 found that the healthy donor cells were mostly normal but the adenomyosis-derived cells commonly had chromosomal abnormalities, particularly aneuploidy and more frequent nonrandom breaks involving chromosomes 7 and 11. The authors report that despite karyotypic instability, the adenomyosis-derived eMSCs stopped cycling after about 26 passages and entered replicative senescence, indicating the observed abnormalities were not associated with transformation/immortalization in vitro.
