Focal Adhesion Kinase-Mediated Sequences, Including Cell Adhesion, Inflammatory Response, and Fibrosis, as a Therapeutic Target in Endometriosis
This study investigated the role of Focal Adhesion Kinase (FAK) in cell adhesion, inflammation, and fibrosis in endometriosis and found that FAK inhibition reduced lesion size and markers of inflammation and fibrosis in a mouse model.

The study investigated focal adhesion kinase (FAK)–mediated links between cell adhesion, inflammation, and fibrosis in endometriosis by analyzing human normal endometrium and endometriotic cyst tissues for FAK and MCP-1, macrophage markers (CD68, CD163), and TGF-β1, and by testing mechanistic effects in immortalized endometrial stromal cell lines (ESC, eESC, and endometriotic cyst–derived CSC). It found higher FAK and MCP-1 expression in endometriotic cysts than in normal endometrium, with increased M2 macrophage infiltration and colocalization of TGF-β1, alongside in vitro evidence that CSC secreted more MCP-1 (enhanced by fibronectin) and that FAK inhibition or JNK inhibition suppressed MCP-1, while FAK inhibition suppressed TGF-β1–driven αSMA induction. In an endometriosis mouse model, a FAK inhibitor reduced lesion number and surface area, decreased Ki67-positive proliferative epithelium, lowered MCP-1, TGF-β1, αSMA, periostin, and reduced F4/80+ macrophage numbers. The main caveat is that the work is largely preclinical and mechanistic, using selected immortalized stromal and macrophage-like systems and assessing outcomes primarily via expression markers and lesion metrics. This paper is centrally about endometriosis — it evaluates FAK as a therapeutic target linking adhesion, inflammatory MCP-1 signaling, and TGF-β1–associated fibrotic changes.
