LIM Kinase 1 Mediates Estradiol Effects on the Phosphorylation of Cofilinl in Eutopic Endometrial Stromal Cells During the Invasion and Proliferation of Endometriosis
Estradiol increases cofilin1 phosphorylation via LIMK1 in endometriosis stromal cells, promoting invasion and proliferation.

The study investigated how estradiol influences invasion and proliferation of eutopic endometrial stromal cells in women with endometriosis by examining the LIMK1/cofilin1 pathway and cofilin1 phosphorylation. Primary eutopic endometrial stromal cells from patients with endometriosis were cultured without estradiol, then estradiol was used to assess phosphorylation of cofilin1 and LIMK1 by western blotting; LIMK1 knockdown was performed using RNA interference, with invasiveness and proliferation measured after silencing. Estradiol increased phosphorylated cofilin1 and LIMK1 levels without changing total cofilin1 or LIMK1 protein, and LIMK1 silencing reduced estradiol-mediated cofilin1 phosphorylation along with decreased invasiveness and proliferation. The paper’s limitation is that it focuses on in vitro primary cell behavior and estradiol/knockdown effects in a pathway rather than validating effects in vivo. This paper is centrally about endometriosis—specifically, estradiol-driven LIMK1-mediated phosphorylation of cofilin1 affecting invasion and proliferation in eutopic endometrial stromal cells.
