The Use of Antibody Arrays in the Discovery of New Plasma Biomarkers for Endometriosis

This study aimed to discover new noninvasive plasma biomarkers for endometriosis using antibody array screening, analyzing pooled plasma samples (n=103 individuals total; 68 endometriosis-positive and 35 negative) stratified by disease status, cycle phase, stage, and phenotype. Using L-series 1000 and Quantibody 660 arrays, the authors screened for differentially expressed proteins, then technically verified ten preselected markers with a custom 10-plex multiplex immunoassay and later by single ELISA; due to limited reproducibility of the L-series 1000 assay, Quantibody 660 was used for primary screening, yielding 280 upregulated and 29 downregulated proteins in the endometriosis pool versus controls. Only four proteins (CD48, DNAM-1, IL-31, XIAP) were confirmed as differentially expressed between pooled samples, and among individually measured initial samples, only IL-31 showed a univariate statistical difference.
