This paper investigated the cyclic (phase-dependent) expression of estrogen receptor beta (ERβ) and multiple ERβ splice variants, alongside ERα, in human endometrium obtained prospectively from women with and without endometriosis using RNA extraction and quantitative RT-PCR. In endometrial samples from healthy (n=10 across two cycle halves) and endometriosis patients (n=8), ERα and ERβ1, ERβ2, and ERβ5 were detected in all cycle phases, whereas ERβ4, ERβΔ125, and ERβΔ1256 were not expressed, with no significant cycle-phase differences in receptor expression overall. Differences emerged between groups, including a significantly higher ERα/ERβ1 ratio in healthy women versus endometriosis patients in the first cycle half, and distinct phase-dependent changes in the relative expression among ERα and ERβ isoforms/splice variants within the endometriosis group; the study is limited by small sample sizes. This paper is centrally about endometriosis — it compares cyclic ERβ splice variant expression and ERα/ERβ ratios in endometrium from patients with versus without endometriosis.
