A dissertation evaluated whether intrauterine administration of CXCL12 or multipotent mesenchymal stromal cells (MSC) from GFP+ donors improves pregnancy rates and placental characteristics in C57BL/6 mice with surgically induced endometriosis, compared with sham-operated controls. After endometriosis induction and 4 weeks of recovery, females received intrauterine CXCL12, intrauterine MSC, or placebo, then were mated and assessed near term by cesarean section, with placentas and adjacent uterine tissue collected from pregnant animals and uterine horns collected from infertile ones; the study explicitly noted that immunofluorescence did not detect MSCs in placenta–decidualized endometrial complexes. Pregnancy outcomes showed 100% gestation in sham groups and in endometriosis groups treated with CXCL12 or MSC (p=0.0168), whereas placebo-treated endometriosis mice had 50% gestation, but MSCs were not found at the analyzed site, suggesting indirect effects via local factor secretion or unmeasured molecular pathways. This paper is centrally about endometriosis — it tests whether intrauterine CXCL12 or mesenchymal stromal cells rescue fertility and affect placental development in an induced endometriosis mouse model.
