Humane endometriale Stromazellen wurden aus Hysterektomie-Präparaten isoliert. Mit 1μM Progesteron und 30 nM Östradiol erfolgte die Dezidualisierung in vitro. An den Tagen 0, 6 und 9 wurden die Zellen für 24h mit rekombinantem hCG (0–100 U/ml) inkubiert. Die mRNA-Expression von IGF-1, -2 und IGF-1R wurde mittels real-time RT-PCR bestimmt. IGF-1 und -2 wurden anhand von ELISAs gemessen, IGF-1R mittels Western Blot detektiert. Im Dezidualisierungverlauf zeigte sich eine rasche und ausgeprägte Steigerung von IGF-1 (Tag 3: 220±26%, p<0.05). Bereits am Tag 6 sank die Expression jedoch wieder ab. IGF-2 war an den Tagen 3, 6 und 9 im Vergleich zu Tag 0 konstant reduziert. IGF-1 konnte am Tag 6 durch hCG reduziert werden (1 U/ml, 77±6%; p<0.05). Unter dem Einfluss von hCG war eine verminderte Expression des IGF-1R in dezidualisierten Stromazellen zu sehen. Die hCG-Inkubation bewirkte jedoch keine Änderungen von IGF-2. IGF-1 und -2 verhalten sich während der Dezidualisierung gegenläufig. IGF-1 zeigt eine ausgeprägte Steigerung in der Frühphase der Dezidualisierung, IGF-2 ist im gesamten Zeitverlauf supprimiert. Möglicherweise ist eine starke Expression von IGF-1 in der frühen sekretorischen Phase für den Beginn der endometrialen Differenzierung von Bedeutung. Der reduzierende Einfluss von hCG auf IGF-1 könnte auf die Notwendigkeit einer niedrigen IGF-1-Expression während des Implantationsfensters hindeuten. Die niedrige Expression von IGF-1R in dezidualisierten Stromazellen und die Reduktion durch hCG würde diese Hypothese unterstützen.
