The paper investigates how human chorionic gonadotropin (hCG) regulates expression of insulin-like growth factor binding protein-1 (IGFBP-1) and tissue inhibitor of matrix metalloproteinases (TIMP)-3 in decidualized human endometrial stromal cells. Endometrial stromal cells isolated from hysterectomy specimens were decidualized with progesterone and estradiol for nine days, then incubated with recombinant hCG for 24–48 hours, with IGFBP-1 and TIMP-3 measured by real-time RT-PCR (and IGFBP-1 also by ELISA), and LH/hCG receptor presence assessed by Western blot; a stated limitation is the in vitro cell model derived from endometrial samples rather than in vivo implantation biology. Decidualization increased IGFBP-1 and TIMP-3 over time, but hCG reduced IGFBP-1 at both mRNA and protein levels and lowered TIMP-3 mRNA, with effects that were dose- and time-dependent. Relevance to endometriosis: the study is about hCG-mediated regulation in endometrial decidualization and implantation-related markers within endometrial stromal cells, which are central to endometriosis-associated endometrial dysfunction, though the paper does not explicitly discuss endometriosis or adenomyosis.
