This study examined cyclooxygenase-2 (COX-2) expression in human endometriosis lesions from three clinical forms—peritoneal (n=76), ovarian (n=42), and rectovaginal (n=8)—using immunohistochemistry and western blot analyses, along with primary and immortalized endometriosis cell cultures/lines. COX-2 expression was quantified and correlated with rASRM stage and biomarkers including ER, PR, and Ki-67, with statistical testing using t-test, Mann–Whitney, chi-square, and Fisher’s exact tests. The results showed varying epithelial COX-2 levels (negative, weak, moderate, or strong), with strong epithelial COX-2 more frequent in ovarian and rectovaginal lesions, while stromal COX-2 was generally low and no strong stromal expression was detected; western blots identified a COX-2–specific 72 kDa band in cell models.
