The paper evaluated the antithrombotic effects of danazol by measuring serial plasma changes in coagulation, fibrinolysis, and platelet-related factors in four patients with endometriosis, and by also administering long-term danazol to a patient with chronic consumption coagulopathy associated with a dissecting aortic aneurysm. Danazol increased regulatory anticoagulant factors including antithrombin III, heparin cofactor II, protein C, and free protein S, while increasing factor II and X and decreasing fibrinogen; among fibrinolytic factors, only plasminogen showed a significant increase. Platelet factors such as platelet counts, ADP-induced aggregability, von Willebrand factor, and thromboxane B2 production showed no consistent changes, and in the coagulopathy patient fibrin degradation products and fibrinopeptides decreased without thrombo-hemorrhagic phenomena, though the study is limited by its very small sample size and includes a single long-term case. Relevance to endometriosis: endometriosis is directly studied in four patients as the main population for the serial factor measurements, though the paper also includes a separate long-term case of coagulopathy related to aortic aneurysm.
