Abstract

Adenomyosis (AM) commonly affects women of reproductive age and can impair female fertility, yet its diagnosis and pathogenesis remain challenging. We compared serum IgG1 Fc‑glycosylation profiles between AM patients (n = 30) and non‑AM controls (n = 16). IgG concentrations were quantified, followed by liquid chromatography‑mass spectrometry (LC‑MS) based comparative analysis of IgG1 glycoform differences. Distinct IgG glycoform variations emerged between the two groups. The AM cohort exhibited a significant reduction in bisecting N-acetylglucosamine( GlcNAc) modifications alongside elevated sialylation levels relative to controls. Furthermore, we performed tissue staining on adenomyosis tissues using lectin and found that the bisecting glycans in the lesion tissue were also lower than those in the control group. To our knowledge, this study provides the first comprehensive characterization of serum IgG glycosylation profiles in AM. The identified alterations in bisecting GlcNAc levels offer novel molecular‑level insights into the pathophysiology of AM and hold promise as potential diagnostic signatures.
