This study examined the effects of Moutan Cortex extract (MCE) on human endometrial stromal cells (HESCs), using assays for viability/proliferation, apoptosis, and cell migration/invasion, alongside Western blot or ELISA to assess signaling pathway mediators. MCE treatment decreased HESC viability and proliferation while increasing apoptosis, and it reduced OPN-induced migration and invasion; OPN also increased osteopontin-driven MAPK signaling and matrix metallopeptidase 9 (MMP9) activation. Overexpression or upregulation of osteopontin reversed the inhibitory effects of MCE, indicating that MCE acts upstream of OPN/MAPK-dependent MMP9 activation.
