This study examined whether peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) regulates aromatase-driven local estrogen biosynthesis in endometriosis. Immunohistochemistry and real-time PCR in endometriosis-related ovarian cyst tissues, compared with eutopic endometrium and myoma endometrium, showed higher PGC-1α expression in endometriosis stromal cells, aromatase expression only in endometriosis tissue, and a significant correlation between PGC-1α and aromatase mRNA. In primary cultures, forced PGC-1α overexpression increased NRHS-dependent luciferase activity, aromatase and SF-1 mRNA, and aromatase activity in endometriosis stromal cells but not in endometrium stromal cells; exon I/promoter usage by aromatase appeared unchanged, and TNF-α—but not estradiol or hypoxia—upregulated PGC-1α.
