Abstract

objective:To investigate effects of DMPA(Depot medroxyprogesterone acetate)on the expression of RANTES(regulated on activation, normal T cell expressed and secreted)in experimental endometriosis models of rats. Methods:Jones’method was used to develop an animal model of endometriosis in rats.Successful models were randomized into the control group and the DMPA-treated group.RANTES concentrations in eutopic and ectopic endometrium and serum were determined by ELLSA.Results:The levels of RANTES in eutopic and ectopic endometrium and serum from DMPA treated rats were 89.2,199.4 and 6 871.5 ng/L respectively(median). In the control group, the concentrations of RANTES in eutopic and ectopic endometrium and serum were 141.8,401.8 and 7 523.3 ng/L respectively(median). The difference of RANTES,between the treatment group and the control group, either in eutopic or in ectopic endometrium, was significant(P0.05 and P0.01,respectively).There was no statistic difference between that in the two groups in serum(PO.05).Conclusion:These studies indicate that DMPA down- regulates endometrial RANTES expression in experimental endometriosis models. The clinical effectiveness of DMPA treatment in endometriosis-associated pelvic pain may be attributed to its inhibition of RANTES production and its suppression of inflammatory responses in the pelvis.
