The variety of expressions of microvessel density, vascular endothelial growth factor and vascular endothelial growth factor receptor (KDR) in endometrium before and after transcervical resection of endometrium

Abstract
Objective To compare the expressions of microvessel density (MVD), vascular endothelial growth factor (VEGF) and vascular endothelial growth factor receptor (KDR) in endometrium before and after transcervical resection of endometrium (TCRE). To investigate the molecular pathogenesis of abnormal uterine bleeding and molecular mechanism of TCRE. Methods All endometrial specimens were in the mid-late-proliferative phase. Ten specimens of simple hyperplasia were collected 12-15 days after curettage and ten of adenomyosis endometrium before TCRE, both of which were regarded as specimens before TCRE. Eight of simple hyperplasia and seven of adenomyosis endometrium were obtained after TCRE, and eight normal endometrial specimens served as controls. MVD, VEGF and KDR were measured immunohistochemically (LSAB). Results The MVD,VEGF and KDR staining indices before the surgery in both simple hyperplasia and adenomyosis groups were significantly higher than those after TCRE and those in normal controls ( P 0 01). But after the surgery it was just same as that of the control. The MVD,VEGF and KDR staining indices between postoperative endometrium and normal ones, simple hyperplasia and adenomyosis, showed no statistic difference, although the mean values of adenomyosis were higher than those of simple hyperplasia. Conclusions Over-expressions of MVD?VEGF and KDR in endometrium, which result in angiogenesis and abundant blood vessel net, are related to abnormal uterine bleeding in simple hyperplasia and adenomyosis. TCRE can down regulate MVD, VEGF and KDR expression in the endometrium with simple hyperplasia and adenomyosis. Adjuvant anti-angiogenesis drugs used after TCRE may be helpful to prevent from recurrence of adenomyosis.
