Abstract
Objective: To establish endometriosis(EMS) infertility in mice models, observe and compare the difference of expression of chemokine EOTAXIN and its receptor CCR3 between endometriosize pregnant and infertile mice, analyze the effect of EOTAXIN/CCR3 gene expression on endometriosis infertility, and to explore Traditional Chinese Medicine Erling capsule intervention. Methods: Established an EMS mouse model, while subcutaneous injection of diethylstilbestrol.Observed after 24 weeks, mice were mated within the same cage, the rate of infertility was calculated and pregnant mice and infertile mice were distinguished.Infertile mice were given oral Erling capsules for treatment for 4 weeks, endometrial in each mouse was drawn and quantitative PCR was used to detect the difference expression of Eotaxin/CCR3 within normal mice, pregnant mice, and infertile mice treated with Erling capsules or not. Results: Eotaxin expression and CCR3 expression were lower in Sham group of mice.Endometrial Eotaxin/CCR3 gene expression was significantly increased in EMS pregnant mice.Eotaxin and CCR3 gene expression in EMS infertile mice is more obvious than in the pregnant mice endometrium. As for infertile mice,Eotaxin/CCR3 expression in mice given Erling capsule was decreased compared to the mice treated with water. Conclusion: EOTAXIN/CCR3 may be directly involved in the pathological process of infertility which caused by endometriosis.Erling capsules can relieve inflammation and improve endometriosis, the mechanisms may be related to directly down-regulated the expression of EOTAXIN/CCR3 in mouse endometrium.
