Abstract

Objective:To study the promotion effect of cAMP on decidualization process in vitro of stromal cells in eutopic endometrium of patients with endometriosis,understand the decidualization ability of eutopic endometrium of patients with endometriosis and study the relationship between endometriosis and infertility as in vitro models of endometriosis. Methods:The eutopic endometrial stromal cells of patients with endometriosis were divided into three groups randomly: non-intervention group,estradiol and progesterone group(the cells were treated with 10 nM estradiol and 1 μM progesterone for 24,48 and 96 hours),estradiol+progesterone+8-Br-cAMP group(the cells were treated with 10 nM estradiol,1 μM progesterone and 0.5 mM 8-Br-cAMP for 24,48 and 96 hours);the levels of prolactin mRNA and insulin-like growth factor binding protein 1(IGFBP1) mRNA in the three groups were detected after collecting the cells. Results:In estradiol+progesterone+8-Br-cAMP group,after treating the cells for 96 hours,the cell morphology changed obviously,the levels of prolactin mRNA and IGFBP1 mRNA in estradiol+progesterone+8-Br-cAMP group were significantly higher than those in estradiol and progesterone group. Conclusion:8-Br-cAMP combined with estradiol and progesterone can promote the decidualization process in vitro of stromal cells in eutopic endometrium of patients with endometriosis obviously,which may be the optimum method of establishing decidualization model in vitro of endometriosis.
