NAT2 and CYP1B1 genetic polymorphisms in patients with genital endometriosis depending on tolerability of melatonin

This study analyzed NAT2 and CYP1B1 gene polymorphisms in 59 women with externally genital endometriosis (stages I–IV confirmed laparoscopically and histologically) who received melatonin (6 mg daily) for 2–6 months, comparing genotypes between those who tolerated melatonin well (64%) and those with poor tolerance who discontinued therapy due to side effects such as drowsiness, frequent nocturnal awakenings, headaches, and insomnia. Using PCR-RFLP on peripheral blood lymphocyte DNA, the authors found that the NAT2 wild-type N allele was more frequent in the poor-tolerance group (38.1% vs 17.1%; p=0.02). They also reported that the NAT2 rapid acetylator phenotype (N/N) together with low CYP1B1 catalytic activity (C/C) occurred more often among poor-tolerators than good-tolerators.
