Abstract

Objective To establish a human endometriosis-derived immortalized eutopic endometrium stromal cell line. Methods The cells from human endometriotic eutopic endometrium were transfected with plasmid PCD2SV40T.Then the cells were screened by G418 and isolated subclone by limited dilution. After that, the subclone was identified by karyotype analysis,immunocytochemistry,RT-PCR,cell doubling time test, nude mice test. Results The established cell line named hEM15A is the first human endometriosis-derived immortalized eutopic endometrium stromal cell line.It revealed the characteristics of endometrium stromal cells. It can be passaged continuously and its clonal structural or numerical chromosome revealed no abnormalities. No tumorigenicity was observed. Conclusion hEM15A cell line is easily accessible and cultured, which could be potentially useful for further research of endometriosis.
