Abstract
Objective: To explore the effect of soybean isoflavone(SIF) on expressions of CD34 and aromatase of rats with endometriosis. Methods: The rat model of endometriosis was established after surgery, then the volume of grafts was measured after three weeks. The rats were randomly divided into blank control group(group Ⅰ),low- dose SIF group(group Ⅱ,0. 5 mg·kg- 1·d- 1),and high dose SIF group(group Ⅲ,50 mg·kg- 1·d- 1); after establishing rat model successfully, the rats were treated with subcutaneous injection of the above- mentioned drug for 12 weeks, then the change of volume of grafts was measured again, the pathological change of transplanted endometrium was observed by HE staining, immunohistochemical SABC method was used to detect the expressions of CD34 and aromatase in endometrial tissues of rats. Results: The successful rate of model establishment was 76. 4% in the study. There was no statistically significant difference in the volume of grafts between before and after treatment in group Ⅱ(P 0. 05); the volume of grafts after treatment was statistically significantly less than that before treatment in group Ⅲ,which was also statistically significantly less than those after treatment in the other groups(P 0. 01). The positive expression rates of CD34 and aromatase in group Ⅱ and group Ⅲ were statistically significantly lower than those in group Ⅰ(P 0. 05),and the positive expression rates of CD34 and aromatase in group Ⅲ were statistically significantly lower than those in group Ⅰ and group Ⅱ(P 0. 01); there was a positive correlation between CD34 expression and aromatase expression in rats with endometriosis(r = 0. 782,P 0. 01). Conclusion: The degeneration effect of SIF on ectopic endometrium is significant in rats, the mechanism may be correlated with decrease of CD34 and aromatase expressions, weakening of migration and growth of endometrial glands and stroma by SIF.
