Deploying host resistance from potato varieties is a promising and complementary strategy. However, the effectiveness of single late blight resistance genes ( Rpi genes) against EU43 and other prevalent clonal lineages remains unclear. In this study, we evaluated the resistance of 9 transgenic potato clones each carrying a single Rpi gene against a panel of 28 P. infestans isolates belonging to the EU43, EU41, EU39, EU13, EU36, and “other” P. infestans clonal lineages. We performed detached-leaf disc assays in the laboratory and whole-plant assays in the greenhouse to compare disease incidence and severity. Our results showed that several Rpi genes (notably Rpi-vnt1.1, Rpi-blb2 , and R9a ) conferred broad resistance against most of the tested isolates, including EU43, whereas most EU43 isolates overcame the R3a and Rpi-blb3 genes. Overall, EU43 did not exhibit significantly greater aggressiveness than other lineages on susceptible hosts. Some EU43 isolates demonstrated a broader virulence spectrum than the EU43 isolates, suggesting that virulence was acquired through mutation, driven by a selective advantage in hosts with the corresponding Rpi gene. However, some EU43 isolates showed a consistently higher virulence and aggressiveness than others.
