Evaluation of Some Safety and Technological Properties of Yeast Strains Isolated from Aliha for Starter Culture Development.

Abstract

Fermentation remains one of the most widespread food preservation and transformation methods, with profound cultural, nutritional, and economic significance, particularly in Africa. Aliha is a traditional Ghanaian maize-based beverage, which is spontaneously fermented and valued for its cultural and traditional benefits in rural and peri-urban communities of Ghana. However, concerns about safety and quality attributes make upscaling difficult and unattractive. This study aims to select yeast strains with the best safety and technological potential for safe Aliha production and potential industrialization. To select the best yeast strains for industrial fermentation of Aliha , the yeast strains need to be assessed for their technological properties, including enzymatic activities (proteolytic, lipolytic, amylolytic, esterases, and β-glucosidases), and survival in acetate and lactate. The yeast cultures were also evaluated on their ability to overcome harsh intestinal environments like 3% bile salts, pH 2, pancreatic juice, pepsin, lysozyme, hemolysis, and screened for antimicrobial resistance. The results illustrate that Saccharomyces cerevisiae JYC2599, Pichia kudriavzevii CBS 5147 18S, and Pichia kudriavzevii CY902 were the best enzyme producers, depicting the highest recovery rates in harsh gastrointestinal conditions, inhibiting most pathogenic organisms, and resisting most antibiotics. They were all hemolytic negative (ℽ), which means they will not have any adverse effects on the host when they are considered as starters for food fermentation. Since poorly or uncharacterized microbial strains can pose health risks and limit the potential for product industrialization and commercial production, the above-qualified yeast strains will help produce Aliha with better qualities, leading to industrialization of the beverage.
