The screening of 139 japonica rice germplasm resources identified seven high-RS varieties for subsequent detailed analysis. The findings revealed that OsGBSSI expression and enzyme activity were significantly upregulated during the early and middle grain-filling stages in high-RS varieties, thus underscoring its crucial role in RS synthesis. In contrast, OsBEIIb expression and enzyme activity exhibited a wave-like pattern that indicated a complex regulatory function in balancing amylopectin and amylose synthesis. Finally, while OsGCD mRNA expression showed no significant correlation with RS content, its enzyme activity was consistently higher in high-RS varieties during the late grain-filling stage, hence suggesting a potential indirect role in RS synthesis. This study provides valuable insights into the molecular mechanisms governing RS accumulation in japonica rice and provide a theoretical basis for breeding high-RS rice varieties with enhanced nutritional quality.
