Abstract
Objective:To determine the dysregulation of micro RNA-556-3p expressions in eutopic and ectopic endometria from endometriosis patients as well as their relationships with the levels of VEGF protein. Method:Fifteen women with endometriosis were recruited as the study group,whose eutopic endometria,ovarian and peritoneal endometriotic lesions were excised. Endometrial tissues from sixteen women without endometriosis were collected as the control group. Taqman real-time PCR was performed to detect the expression levels of mi R-556-3p in the above groups,and Western blot was used to examine the protein levels of VEGF. Moreover,the correlation of VEGF protein with mi R-556-3p levels was analyzed. Results:Compared with the control group,the expressions of micro RNA-556-3p in eutopic endometria and peritoneal lesions were significantly elevated(P0.001),with statistically significant decreases in VEGF protein levels in all the eutopic or ectopic endometrial tissues(P0.001,P=0.02,P0.001 respectively). Besides,the ovarian endometriotic lesions had a higher level of mi R-556-3p than eutopic endometrial(P0.001). Whereas,the downregulation of mi R-556-3p and overexpression of VEGF protein were observed in peritoneal lesions compared with the eutopic endometrial tissues(P0.001). Moreover,the levels of mi R-556-3p were inversely correlated with the VEGF protein levels in both eutopic endometria and peritoneal endometriotic lesions(r=-1.026,P=0.01;r=-1.819,P=0.04,respectively). Conculsion:Dysregulation of mi R-556-3p might cause the abnormal expression of VEGF and contribute to the pathogenesis of endometriosis.
