Abstract

Objective:To investigate the effects of Danazol on ectopic endometrium and the effect of Danazol on expression of IL-6 mRNA in the ectopic endometrium.Methods:To establish the rat models,Danazol was administered repeatly as a s.c.injection per day separately for 2、4 、6 and 8 weeks(n=29).The control groups(n=6) were injected s.c.with normal saline.After treatment,the laparotomy was performed again to investigate the state of the endometrial explants.Endometriotic tissue was cut from group treated with Danazol for eight weeks and control group.IL-6 mRNA expression in the ectopic endometrium was examined by reverse transcription-polymerase chain reaction(RT-PCR).Results:The 3rd week after modelling,no significant difference was noted for the size of explants between groups(P 0.05).After treatment with Danazol separately for 2、4、6、and 8 weeks,the volume of endometrial explants was reduced significantly(P 0.01),especially 4、6、8-week administered groups(P0.01).The 2nd week after treatment,ectopic endometrium was not further proliferated and the 4th、6th、8th weeks after treatment,the tissue became atrophy.8 weeks after treatment,the level of IL-6 mRNA reduced significantly compared with that of control group(P0.01).Conclusion:Danazol treatment can induce ectopic endometrium atrophy gradually;its effect correlates with treatment duration.Danazol treatment can decrease the level of IL-6 mRNA in endometriotic tissue significantly.
