Abstract

Abstract To investigate and understand the Feline Coronaviruses infection in South China tigers, and to provide a basis knowledge for monitoring and prevention the disease, 119 whole blood and serum samples from South China tiger in 11 zoos were analyzed for aetiology and serology using PCR and ELISA methods. The results indicated that no Feline Coronaviruses antigen was detected in 119 whole blood samples via PCR. 19 positive samples were detected by ELISA in serum samples. The total seropositive rate was 15.97%. The positive rate of antibodies across locations varied from 0% to 66.67%, with a median of 11.11%. Seroprevalence rates varied significantly among locations (P = 0.000); the juveniles exhibited an seroprevalence rate of 17.39%, while adults had a rate of 15.63%, with no significant difference observed(P = 0.455); positive rates were 17.24% for males and 14.75% for females. There was no significant difference observed in the seroprevalence rate of Feline Coronaviruses between sexes (P = 0.207). The seroprevalence of Feline Coronaviruses in South China tigers was contributed by zoos (different locations).

South China Tiger Feline Coronaviruses RT-PCR ELISA Introduction Following the COVID-19 outbreaks, significant attention has been focused towards coronavirus, epidemiological studies, diagnostic methodologies, and preventive strategies (Du et al., 2021). Veterinary medicine has extensive knowledge of coronavirus diseases, given the variety of coronavirus species (CoV) impacting wild animals, many of which remain unidentified (Paltrinieri et al., 2020). Several surveys of feline coronaviruses (FCoV) reveal that FCoV colonisation of the alimentary tract is common in majority feline populations (Ouyang et al., 2022 ). The feline coronavirus, belonging to the coronaviridae family, is capable of causing intestinal infectious diseases in cats. The primary symptoms consist of fever, vomiting, diarrhea, and loss of appetite. Severe cases can lead to dehydration, predominantly affecting young felines. The primary mode of transmission for FCoV is fecal-oral transmission. FCoV infects enterocytes in the small intestine via fecal-oral transmission, replicates locally, and is subsequently released in fecal samples.(Pedersen,et al,.2008). Cats can excrete considerable amount of virus via their faeces post-infection, alongside the virus persisting in the faeces for several weeks (Felten et al., 2020; Kipar Anja et al.,2010; Hartmann Katrin, 2005 ).Feline coronavirus is a highly contagious virus that is globally prevalent. The pathogen can be classified into two distinct biotypes according to their pathogenicity profiles: Feline Infectious Peritonitis Virus (FIPV) and Feline Enteric Coronavirus (FECV)(Herrewegh, et al., 1998; Haijema et al., 2004 ). Research indicates that the feline infectious peritonitis virus is a mutation of the feline intestinal coronavirus, exhibiting greater pathogenicity (Pedersen et al., 2009). FCoV is assigned to two distinct serotypes, FCoV I and FCoV II, based on antigenicity and the sequence of the S gene(Gao et al, 2023). FCoV affects up to 90% of cats worldwide (Jähne et al, 2022) .Cats serve as natural reservoirs for feline coronavirus. The zoo environment facilitates their survival and proliferation, leading to ecological niche overlap with captive wildlife. This proximity may promote FCoV transmission through direct bodily fluid contact or fecal contamination, posing a potential threat to wildlife population health and biosecurity systems‌. The South China tiger ( Panthera tigris amoyensis ), a large unique feline in China. As of 2025, zoos nationwide house over 200 tigers (as of 2024), indicating a concerning population status. Further research is necessary to understand the susceptibility of the South China tiger to feline infectious diseases. This study investigates the epidemic situation of the Feline coronavirus in the South China tiger population. The study collected 119 blood samples of South China tigers across different zoos and investigated the feline coronavirus infection throughout the South China tiger population applying molecular biology and serological methods. The objective was to learn the infection of feline coronavirus in South China tiger within the zoo and provide essential data for disease prevention and control. Materials and methods Sample collection Blood samples (n = 119) from captive South China tigers were sourced from 11 zoos in China (Table 1 ): Shanghai Zoo, Nanchang Zoo, Chengdu Zoo, Suzhou Zoo, Hangzhou Zoo, Luoyang Zoo, Chongqing Zoo, Meihuashan Nature Reserve, Guangzhou Zoo, Zhengzhou Zoo and Linyi Zoo. The animals were anesthetized with ketamine (3–5 mg/kg) and xylazine (2 mg/kg) administered intramuscularly after a 12-h fast. This anesthesia protocol minimizes physiological stress on animals, ensuring the stability of conditions. Blood samples (3–5 mL) were collected via venipuncture and immediately dispensed into tubes containing either EDTA-K2 anticoagulant or serum separator gel.Whole blood samples were centrifuged at 3,000 g for 5 minutes to separate serum, and the resulting serum samples were stored at -80°C for downstream analysis. Polymerase chain reaction (PCR) was performed in a final volume of 20 µL, with the concentration of each component adjusted according to the recommended reaction system of the kit. The annealing temperature was set to 55°C. PCR products were analyzed by electrophoresis in 1.5% agarose gels, and the results were observed and recorded by a gel imaging system(Shanghai Tanon Life Science Co.,Ltd). ELISA screen The presence of serum antibodies against feline coronavirus was detected using the EVL FCoV test ELISA kit (European Veterinary Laboratory, Woerden, Netherlands), with the procedure performed according to the manufacturer's instructions provided in the following section: All kit components and samples are equilibrized to the room temperature prior to testing. Positive and negative controls were diluted to a ratio of 1:30. The serum samples were diluted to 1:150 and assayed in duplicate. Absorbance values were read immediately at 450 nm using an ELISA plate reader(Mindray,MR-96A,China). The positive control was required to exhibit a mean optical density (OD) value ≥ 0.8, whereas the negative control must have a mean OD value < 0.4. The S/P ratio was calculated using the formula S/P = (OD 450 of the sample - OD 450 of negative control)/ (OD 450 of positive control - OD 450 of negative control). Samples with S/P ratios of 0.24 or greater were considered as positive for feline coronavirus antibodies, whereas S/P values below 0.24 were considered negative. Data analysis Using SPSS (version 23.0, SPSS Inc., Chicago, IL, USA), the Chi-square test was employed to analyze the prevalence of feline coronavirus infection in South China tigers across sex, age, and region. The Kruskal-Wallis test was applied for data from various regions. The Mann Whitney U test was applied for data differentiated by sex and age. A probability value (p-value) of less than 0.05 indicated statistically significant. Results Dectection of FCoV in SCT blood samples The total of 119 whole blood samples were negative for FCoV PCR detection. Seroprevalence of FCoV antibody in South China tigers 119 serum samples of South China tiger from 11 zoos were tested for feline coronavirus antibody via ELISA. The results indicated that 19 of the 119 serum samples of South China tiger were positive, with a positive rate of 15.97%. The positive rate of feline coronavirus antibody in Guangzhou Zoo was 66.67% as the highest, and the positive rate in Suzhou Zoo was 0.00% as the lowest. There was a significant difference in the serum antibody of South China tigers in different zoos (p < 0.05). Seroprevalences of antibody to feline coronavirus in South China Tiger from different zoos (p = 0.000). Due to the small number of samples from Zhengzhou Zoo and Linyi Zoo, these two zoos were not included in the non-parametric test for regional differences. The positive rate and S/P value of feline coronavirus antibody in different zoos are shown in Table 2 . The difference in FCV antibody seroprevalences between adult and juvenile tigers did not reach the level of statistical significance (p = 0.455). The positive rate of feline coronavirus antibody and the S/P value of South China tigers from different ages are shown in Table 3 . There was no significant difference in seroprevalences of antibody to feline coronavirus in South China tigers of different genders (p = 0.207). The antibody positive rate of feline coronavirus and the S/P value of South China tigers of different genders are shown in Table 4 . Only zoos or other breeding institutions house it in captivity, with a population of less than 200. As an important factor affecting population breeding, infectious diseases pose a threat to the health and survival of the South China tiger, necessitating careful attention. FCoV is prevalent in felines. Cats infected with FCoV may develop Feline Infectious Peritonitis (FIP), a disease with a high case fatality rate. FCoV infection follows a fecal-oral pattern of dissemination, with infection prevalence typically correlated with feline population density and co-housing conditions. Immunosuppressed individuals exhibit heightened susceptibility to developing FIP upon FCoV exposure (Hu et al., 2024). Once widely disseminated, it will significantly impact feline well-being. The living environment is relatively poor. Poor sanitation in the environment is one of the possible causes of infection, and it can also lead to a mutual infection (Drechsler et al., 2011). Due to the body size of South China tiger, there is limited research on FCoV infections. To improve the breeding environment of the South China tiger and diminish the impact and threat of the feline coronavirus on the South China tiger, relevant monitoring and control activities are required, which will also provide a scientific foundation for the prevention of thefeline coronavirus disease. This study investigated 119 samples of nucleic acid and serum from a South China tiger from 11 zoos, looking for signs of feline coronavirus infection. While current nucleic acid testing showed no positive pathogenic samples of feline coronavirus, epidemiological data from Liu Qiujin ( 2019 ) indicate a 72.2% prevalence rate in clinically healthy cats, suggesting potential subclinical viral carriage.This indicates that the South China tiger is in a relatively safe living environment or is less prone to feline coronavirus, possibly due to the individual differences between the species. Furthermore, it is possible that the South China tiger may experience a temporary infection with feline coronavirus within a certain period but will ultimately recover from the virus and will eventually heal itself. Therefore, the absence of a corresponding pathogen does not rule out the susceptibility of the South China tiger to feline coronaviruses. There is still a risk of infection and outbreak due to environmental pollution or other factors. The feline coronavirus infection of the South China tiger in zoos of different regions was significantly different (p < 0.05), which may be caused by differences in the zoos’ environments. It is reported that feline coronavirus infection exists in South China tigers of varying sexes and ages. The S/P value does not reach a significant level (p > 0.05), suggesting that South China tiger individuals of different ages and sexes are susceptible to feline coronavirus. Kittens in cat studies are more susceptible to FCoV (Li et al., 2021; Hong, 2023 ). Moreover, we observed that the FCOV prevalence rate in males was slightly higher than that in females. However, in Dong's study, female cats are more susceptible, potentially due to species differences (Dong et al., 2024). Currently, domestic zoos primarily administer vaccinations to dogs, cats, and other animals. If conditions allow it, they can give cats six or seven vaccines to protect the South China tiger from FCoV infection and keep the population healthy. Brostoff observed that the feline coronavirus mRNA vaccine can elicit humoral immunity and cell-mediated immunity in mouse hosts, which provides a foundation for moving forward with immunogenicity and safety studies in feline species (Brostoff et al., 2024). The study by Chawla applied immunoinformatic tools to characterize immunogenic epitopes within the S-protein of FCoV, culminating in the design of a pioneering vaccine(Chawla et al., 2023). These provide technical support for effective feline immunization in the future. According to research findings, approximately 60% of newly emerging human pathogens are zoonotic, with over 71% of them emanating from wild animals (Cutler et al., 2010). Infectious diseases, such as viruses and bacteria, have numerous sources and routes of infection in the complex environment of the zoo. Certain infectious diseases, including zoonoses, pose a threat to herbivores, carnivores, and primates. In an environment that hosts a large number of tourists, a variety of wild animals, and a complex community structure, it is crucial to understand the source of infection, control the transmission of pathogens, and closely monitor susceptible animals and high-risk host animals(Hu et al.,2023). The capability to accurately predict the transmission of pathogens across species in a timely manner (Qin et al., 2021). If the South China tiger is infected with coronavirus, it is recommended that it be reintroduced into the population at least two months after the South China tiger recovers to reduce the possibility of infecting other animals (Addie et al., 2009). In addition, we should ensure that animal cages have outdoor access. Animals that are without outdoor access are more susceptible to becoming infected (El-Tholoth et al., 2023). We aim to establish a secure and comfortable habitat for the captive rearing of uncommon, wild creatures, which will provide strong assurances for the population's growth. The results indicated that no Feline Coronaviruses antigen was detected in 119 whole blood samples via PCR. 19 positive samples were detected by ELISA in serum samples. The total seropositive rate was 15.97%. The positive rate of antibodies across locations varied from 0% to 66.67%, with a median of 11.11%. Seroprevalence rates varied significantly among locations (P\u0026thinsp;=\u0026thinsp;0.000); the juveniles exhibited an seroprevalence rate of 17.39%, while adults had a rate of 15.63%, with no significant difference observed(P\u0026thinsp;=\u0026thinsp;0.455); positive rates were 17.24% for males and 14.75% for females. There was no significant difference observed in the seroprevalence rate of Feline Coronaviruses between sexes (P\u0026thinsp;=\u0026thinsp;0.207).
