Isolation and Culture and Identification of Human Endometrial Glandular Epithelial Cell and Stromal Cell as an in vitro Model of Endometriosis

Abstract
Objective To establish highly purified culture of endometrial glandular epithelial cells and stromal cells as an in vitro cell model of endometriois. Methods Mixed enzymolysis, serial filtration and gradient centrifugation were used to isolate, purify and culture endometrial glandular epithelial cells and stromal cells in endometrium from woman endometriotic tissues. The cells were identified by microscopy,immunohistochemistry and immumofluorescence. Results The endometrial cells were cultured successfully , the percentages of the cytokeratin positive stromal cells and of the vimentin positive glandular epithelial cells were 90%-95% and more than 90% respectively. Conclusion The endometrial glandular epithelial cells and stromal cells were cultured successfully,can be used as in vitro cell models of endometriosis.
