Effect monoclonal antibody against vascular endothelial growth factor on the transplanted endometriosis lesions in SCID mice

Abstract

Objective To investigate the effects of monoclonal antibody against vascular endothelial growth factor(VEGF) on the growth and angiogenesis of transplanted endometriosis lesions in the severe combined immunodeficiency disease(SCID) mouse model.Methods Eutopic endometrium from women with endometriosis was transplanted to SCID mice,and the mice were randomly divided into test group and control group.After two weeks of transplantation,the test group(n=10) was injected with VEGF monoclonal antibody(3 mg.kg-1.d-1) into the peritoneal cavity.The control group(n=10) was given same volume of sterile PBS(200 μl.d-1).The size of endometriotic lesions in SCID mice was measured every three days,and all the treatment lasted for 14 days.Microvessel density(MVD) and the expression of VEGF were determined immunohistochemically.Results VEGF monoclonal antibody inhibited the growth of endometriotic lesions in mice(P0.05).Microscopic examination showed the bulk of the endometrial tissue decreased and glands depauperated.The glandular epithelium partially degenerated,and necrotic debris was presented in the endometrial stroma.MVD and the expression of VEGF were decreased significantly in the test groups compared to the control group(P0.05).Conclusions Monoclonal antibody against VEGF can effectively interfere the growth of endometriosis by inhibiting angiogenesis.This suggests that the use of angiostatic agents may be promising as a therapy for endometriosis.
