Abstract

Objective To investigate the effect of Triptorelin on the release of tumor necrosis factor-α(TNF-α) and vascular endothelial growth factor (VEGF) in the in-vitro eutopic endometrial cells of adenomyosis.Methods Biopsy specimens of eutopic endometrium obtained from 20 women with adenomyosis and 16 normal women were studied.Cells were cultured with Triptorelin in the concentration of 10-7mol/L and 10-5mol/L. TNF-α and VEGF concentrations in culture supernatant were detected by commercial enzyme-linked immunosorbent assay (ELISA).Results The concentrations of VEGF and TNF-α in the eutopic endometrial cells of adenomyosis were significantly higher than those in the normal endometrium. The concentrations of VEGF、TNF-α in the both groups were downregulated by Triptorelin in a dose-dependent manner. Conclusion VEGF and TNF-α may play an impotant role in the pathogenesis of adenomyosis. Triptorelin can directly suppress the survival and growth of ectopic endometrial by decreasing the release of VEGF and TNF-α which are related to the adenomyosis angiogenesis.
