Study of tissue morphology and vascular endothelial growth factor and matrix metalloproteinase-9 gene expressions in nude mouse endometriosis model

Abstract

目的 建立子宫内膜异位症(内异症)裸鼠模型,并对其相关生物学行为进行研究.方法取人分泌晚期子宫内膜,剪碎,置入裸鼠盆、腹腔不同部位,内异症(EM组)和非内异症(NEM组)裸鼠各24只;对照组:3只裸鼠,同法将人大网膜组织置入裸鼠盆、腹腔不同部位.各组裸鼠分别于置入后第5、15、30天随机脱颈椎处死,观察异位病灶生长情况,并留取异位病灶,在光镜及透射电镜下观察其形态学变化.RT-PCR法检测在位内膜及异位病灶血管内皮生长因子(VEGF)、基质金属蛋白酶9(MMP-9)mRNA表达,免疫组化链霉素抗生物素蛋白-过氧化物酶连接法检测异位病灶雌、孕激素受体表达.结果内膜置入5 d,即见异位病灶牢固黏附,血液供应丰富.光镜下见腺体细胞及散在间质细胞,且随时间延长,异位病灶与裸鼠组织融合越明显.15 d时,电镜下可见异位病灶内炎性细胞增生、浸润最明显,30 d时腺细胞中细胞器消失,但仍可见分泌颗粒,核染色质聚集,并有凋亡趋势.与在位内膜比较,异位病灶VEGF、MMP-9 mRNA表达增强(P 0.05).结论裸鼠作为内异症研究的动物模型,科学、简便、可行,其异位病灶发生、发展过程与人类内异症相似,且内异症是多因素相关性疾病。
