Abstract

SUMMARY: The lack of diagnostic markers of endometriosis causes the surgery to be performed for this purpose. It is an invasive method of high cost contributing to the global scenario, where there has been an average of seven years before the disease is diagnosed from the first symptoms. Objective: This study is based on the changes of gene expression in the endometrium of women with endometriosis. Since the body and cervix have common embryonic origin and receptors for estrogen and progesterone have forum described in the cervix, this study aims to identify whether there are differences in gene expression in endocervical epithelium with the prospect of creating a non-invasive test for the diagnosis of disease. Methods: We performed RT-PCR analysis of the endocervical epithelium of 4 women with deep endometriosis and 6 healthy women. RESULTS: Eight genes showed overexpression in samples of women with endometriosis, five genes of genes related to cell cycle control (CCNB1, CCNG1, CUL1, GTF2H1, PCNA) and three genes inflammatory cytokines (C3, CCL21 and CXCL14). CONCLUSION: Analysis of gene expression in endocervical epithelium of women with endometriosis is a promising non-invasive screening tool for the diagnosis of endometriosis

Trata-se de um método invasivo de alto custo colaborando para o cenário mundial, em que observa-se em média sete anos para que a doença seja diagnosticada desde os primeiros sintomas. Objetivo: Este estudo tem como base as alterações de expressão gênica no endométrio de mulheres com endometriose. Visto que o corpo e colo uterino possuem origem embrionária comum e receptores para estrogênio e progesterona já fórum descritos na cérvix, este estudo visa identificar se há diferenças na expressão gênica no epitélio endocervical com a perspectiva de criação de um teste não invasivo para o diagnóstico da doença. Métodos: Realizamos análise com RT-PCR do epitélio endocervical de 4 mulheres com endometriose profunda e 6 mulheres saudáveis. Resultados: Oito genes apresentaram hiperexpressão em amostras de mulheres com endometriose, cinco genes dos genes relacionados ao controle do ciclo celular (CCNB1, CCNG1, CUL1, GTF2H1, PCNA) e três genes a citocinas inflamatórias (C3, CCL21 e CXCL14). Conclusão: Análise da expressão gênica no epitélio endocervical de mulheres com endometriose é uma promissora ferramenta de rastreio não invasivo no diagnóstico de endometriose.
