The study of selective estrogen receptor-β agonist in the treatment of adenomyosis in mice

Abstract
Objective To investigate the effects of selective estrogen receptor-β agonist(WAY-20007)on the development of adenomyosis(AM).Methods Ectopic endometrium from adenomyosis was subcutaneously transplanted into severe combined immunodeficiency disease(SCID)mice.To establish the AM mice model.The next day the mice were randomized into treatment group(n=26)received WAY-20007(5 mg/kg·d)by subcutaneous injection and the control group(n=25)received normal saline.Lasted 14 days,then measured the weight of the lesions.Immunohistochemistry and RT-PCR were used to detect the expression of ERβ in ectopic tissue.Results The weight of lesion reduced in mice from treatment group comparied with that in the control group(P0.05).Atrophic gland,nonintegrated structure,and necrosis in stroma could be found in the treatment group.Expression of CD10 and CK were decreased significantly in the treatment group than that in the control group(P0.05),while expression of ERβ in the treatment group was no significantly than that in the control group(P=0.1419).Conclusion Selective estrogen receptor-β agonist can interfere with the growth of adenomyosis effectively,which may be by the selective estrogen receptor-β.
