This study confirmed the specificity of p53 staining in endometriosis by using negative controls and blocking endogenous biotin activity.

Abstract

I thank Dr. Mittal for indicating a very important point. We stained parallel sections using normal mouse serum (instead of anti-p53 antibody) as a negative control, and no staining was observed in the sections that showed positive p53 staining. We further examined the possibility of endogenous biotin activity by preincubating the sections with streptavidin (Pierce, Rockford, IL). Because the positive p53 staining was not reduced or lost even after this blocking test, we believe that the positive p53 staining is specific and not due to endogenous biotin activity.
