Gene expression of 15 genes in eutopic endometrium of women with endometriosis, adenomyosis and controls measured by NanoString

Abstract

We aimed to validate 15 putative markers involvement in the development of deep infiltrating endometriosis, ovarian endometriosis, and adenomyosis and evaluate their potential as biomarkers for these three conditions. An RNA-Seq discovery study using endometrial samples identified differentially expressed genes. The validation by Nanostring (n=285) includes 110 controls, 57 adenomyosis, 60 deep infiltrating endometriosis (DIE), and 58 ovarian endometriosis (OE). RNA was obtained from 285 blocks of formalin-fixed paraffin embedded (FFPE) eutopic endometrial samples by using High Pure FFPET-RNA Isolation Kit (Roche) and quality controls were performed. Then, they were analyzed by Nanostring following manufacturer’s instructions. Post-quality controls were also performed by using nSolver analysis software (Nanostring). This dataset includes the raw files from the validation study by using a custom NanoString codeset (RCC files) (n=285). The dataset includes the excel file with the normalized data for each gene studied, which includes 20 genes (15 candidates to validate and 5 houskeeping genes) and 14 internal controls from the manufacturer and one excel file with the metadata of each of the 285 patients.
