GATA6 impairs pancreatic cancer cells stemness by modulating Wnt/β-catenin signaling

Abstract

Background: Cancer cells' stem cell capabilities are thought to have a role in the beginning and development of pancreatic ductal adenocarcinoma (PDAC). The Wnt/β -catenin signalling pathway is an important regulator of cancer cell stemness. Methods: : In this article, through a small range of genetic screening, we found GATA Binding Protein 6 (GATA6), It belongs to a family of transcription factors that attach to the (A/T)GATA(A/G) consensus pattern to stimulate or repress gene expression. It was discovered to be overexpressed in certain PDAC patients with a good prognosis. The biological activities of GATA6 in pancreatic cancer, however, are unknown. In current investigations, we used western blotting assays, qPCR assays, Dual-luciferase reporter assays and Chromatin immunoprecipitation assays in combination with various cytological experiments to explore the molecular mechanisms involved. Results: : Overexpression of GATA6 suppresses pancreatic cancer cell development both in vitro & in vivo by affecting stemness & downregulating the expression of stemness-related markers CD133, ALDH, CD44, & SOX-9, according to the findings. GATA6 upregulates the expression of stemness-related genes by inhibiting Wnt/β-catenin signalling & suppressing β-catenin nuclear translocation. Conclusion: GATA6 inhibits the stemness features of PDAC cells via the Wnt/β-catenin signalling pathway, according to this study. As a result, it might be a viable therapeutic target for cancer therapy.
