Mechanisms of angiogenesis effect of Artesunate on endometriosis of model rats:An experimented study

Abstract
Objective:To explore the mechanism and evaluate Artesunate's effect on endometriosis and angiogenesis by detecting ectopic endometrium volume,VEGF expression and micro vessel density(MVD)in ectopic endometrium of model rats.Method:Wistar rat endometriosis models were established,and after 6 weeks,the successful endometriosis rats were divided into control group,Artesunate 10mg/kg·d group,Artesunate 50mg/kg·d group and Artesunate 100mg/kg·d group,and the rats were treated persistently for 15 days with intragastric administration.The ectopic focus volume was observed,and vescular endothelial growth factor(VEGF) was observed in ectopic endometrium by immunohistochemistry(S-P technique).MVD was determined by immunostaining for CD34.Results:①The volume of grafts of different therapy groups at the third operation was obviously reduced compared with the second operation(P0.05),and was significantly reduced compared with the control group at the third operation(P0.05).②The expression of VEGF and MVD of each therapy group treated with Artesunate was significantly lower than that control group in the same period(P0.05).③With increase in Artesunate,the volume of grafts,VEGF expression and MVD of Artesunate group were significantly lower than those of control group.④There was a significantly linear positive correlation between the expression of VEGF and the MVD of each therapy group treated by Artesunate(P0.05).⑤The positive expression of VEGF and MVD had no correlation in control group(P0.05).Conclusion:Artesunate can decrease the volume of grafts,inhibit the expression of VEGF and reduce the number of MVD in rat ectopic endometriosis,suggesting Artesunate has anti-angiogenesis effect by inhibiting the expression of VEGF,and therefore,Artesunate would be a new medicine of ant-angiogenesis for endometriosis.
