The purpose of this work is to illustrate how Trim59 affects sleep disturbances and the resulting hippocampus damage and cognitive impairment. Methods: PCPA injection was used to create the mouse sleep disorder model, and AAV-Trim59 was stereotaxically injected into the brain to identify Trim59 protein expression in the mouse hippocampal region. Sleep duration and latency were assessed using the pentobarbital sodium sleep experiment, and the hippocampal neurotransmitters 5-HTP, 5-HT, GABA, and Glu were identified using ELISA. The water maze experiment was performed to measure swimming speed, escape delay, number of platform crossings, and target quadrant residence duration. The injury to the hippocampus was examined using H&E staining. The expression of the NLRP3 inflammasome in hippocampal tissue was found using Western blot. Results: Mice who are sleep-deprived exhibit decreased levels of Trim59 expression in their hippocampal regions. Trim59 enhances 5-HTP, 5-HT, and GABA in the hippocampus, decreases Glu content, shortens sleep latency, and extends the duration of sleep in PCPA mice. Additionally, Trim59 lengthens the duration spent in the target quadrant, increases the frequency of platform crossings, and decreases the escape latency of PCPA mice. Trim59 suppresses the expression of the NLRP3 inflammasome protein and lessens hippocampus tissue damage. Conclusion: Trim59 attenuates sleep disorders and enhances learning memory in sleep-deprived mice.
