Effect of 17β-estradiol on expressions of SDF-1/CXCR4 mRNA and protein in eutopic endometrium of patients with endometriosis

Abstract

Objective:To research the effect of 17β-estradiol on expressions of SDF-1 and its receptor CXCR4 mRNA and protein in eutopic endometrium of patients with endometriosis,explore the function of SDF-1/CXCR4 in occurrence and development of endometriosis.Methods:Stromal cells of eutopic endometrium in patients with endometriosis were separated and cultured in vitro,then they were treated by 17β-estradiol of various concentrations(10-12,10-10,10-8,10-6 mol/L) for 12,24 and 48 hours,real-time quantitative PCR(RQ-PCR) and flow cytometry were used to detect the expression levels of SDF-1/CXCR4 mRNA and protein before and after treatment with 17β-estradiol;the effect of estrogen receptor antagonist ICI182780(10-8 mol/L) on expressions of SDF-1/CXCR4 mRNA and protein was analyzed.Results:After treating stromal cells of eutopic endometrium in patients with endometriosis by 17β-estradiol of various concentrations for 12,24 and 48 hours,the expression levels of SDF-1/CXCR4 mRNA and protein increased to varying degrees,the peak appeared when the concentration was 10-10mol/L;estrogen receptor antagonist ICI182780 inhibited the expression of 17β-estradiol obviously.Conclusion:Estrogen can promote the expressions of SDF-1/CXCR4 mRNA and protein in eutopic endometrium of patients with endometriosis,SDF-1/CXCR4 may participate in the occurrence and development of endometriosis.
