Effect of GnRHa on cell proliferation and VEGF's secreting of ectopic and eutopic stromal cells from patients with endometriosis in vitro

【Objective】 To investigate the effect of GnRHa on the cell proliferation and VEGF's secreting of ectopic and eutopic endometrial stromal cells from patients with endometriosis(EMs) in vitro.【Methods】 The ectopic and eutopic endometrial stromal cells were isolated,cultured and identified in vitro,then added to 10-10M、10-8M、10-6M GnRHa.The growth and proliferation of ectopic and eutopic stromal cells were measured by MTT assay;ELISA was used to measure the VEGF concentration change.【Results】 GnRHa can inhibit the proliferation of ectopic and eutopic endometrial stromal cells from patients with endometriosis,in a dose-and time-dependent manner(P 0.05),and the effect on ectopic was stronger than the eutopic(P 0.05);The concentration of VEGF in ectopic and eutopic cells was significantly decreased after the treatment of GnRHa,in a dose-dependent manner(P 0.05),and the ectopic was lower than the eutopic in 10-6M GnRHa(P 0.05).【Conclusion】 GnRHa can significantly inhibit the cell proliferation of ectopic and eutopic endometrial stromal cells of EMs and decrease the secretion of VEGF.
