The expression and significance of four genes on Wnt/β-catenin signal pathway in endometriosis

Abstract
Objective To explore Wnt-7a,Frlzzled,β-catenin and MMP-9genes expression and significance of Wnt/β-catenin signal pathway in endometriosis of situ tissues and malposition tissues and normal endometrial tissues.Methods Wnt-7a,Frlzzled,β-catenin and MMP-9protein expression was detected respectively in endometriosis of situ tissues and malposition tissues and normal endometrial tissues using immunohistochemistry and Western blot method.Wnt-7a,Frlzzled,β-catenin and MMP-9 mRNA expression was analyzed by real time quantitive RT-PCR method.Results By using immunohistochemistry method,there was no Wnt-7aprotein expression in the three different tissues.Frizzled,β-catenin and MMP-9protein expression level in malposition of endometriosis tissues was significantly higher than in normal endometrial tissues(P 0.01);malposition of endometriosis tissues was also significantly greater than in situendometriosis tissues.By using Western blot method,Wnt-7a,Frizzled,β-catenin and MMP-9protein expression in malposition of endometriosis tissues was higher than in normal endometrial tissues and situ of endometriosis tissues.In addition,Wnt-7a,Frlzzled,β-catenin gene mRNA expression level malposition in of endometriosis tissues was higher than in situ of endometriosis tissues and normal endometrial tissues.Wnt-7a,Frlzzled,β-catenin genes mRNA expression in situ of endometriosis tissues was higher than in normal endometrial tissues.However,MMP-9gene mRNA expression level in situ of endometriosis tissues was lower than in normal endometrial tissues.Conclusion The four genes Wnt-7a,Frlzzled,β-catenin and MMP-9on Wntβ-catenin signal pathway participate in pathogenesis of endometriosis.and maybeβ-catenin,through conducting the upstream genes Wnt-7a,Frizzled and downstream signal target gene MMP-9mRNA,acts as a mechanism in the development of endometriosis.
